摘  要:研制并比较了7种不同类型的黄曲霉毒素B1纯化微柱,优选出AFTB1-I亲和微柱,具有分离高效、操作简便适于快速检测的特点。设计出与微柱净化配套使用的半自动化负压装置,可简化净化操作步骤、缩短操作时间。对亲和微柱的进样条件、淋洗条件、洗脱条件、流速、结合容量、加标回收率、稳定性等进行了研究,结果表明:微柱的结合容量〉25μg/kg,不同水平加标回收率范围为80.00%-91.50%。不同批次微柱的加标回收率相对标准偏差为1.76%,无显著差异。同时,采用现有国标方法,利用本实验研制的亲和微柱对实际样品净化,不同水平加标回收率范围为80.00%-92.00%,能满足国标检测限量范围内粮油中黄曲霉毒素B1的快速检测要求。A new micro - column combined with a semiautomatic negative - pressure apparatus for aflatoxin B1 purification characterized by high performance, simplicity and convenience for use was developed from seven selfdesigned micro - columns based on immunoaffinity. The condition such as injection, rinse, elution, flow velocity, binding capability, recovery and stability of the columns were studied and optimized. The results showed that the binding capability of the column exceeded 251μg/kg, and the recoveries rate were from 80.00% - 91.50% in different level. The RSD of different batches were 1.76% which indicated that different batches of columns had no significant differences. According to national standard method, the purifying of aflatoxin B1 in grain and oil samples was carried out by the new self - designed columns, the recoveries were in range of 80.00% - 92.00%. It was concluded that the new column could satisfy the request of national standards for aflatoxin B1 determination in grain and oil.
