反义EGFR寡核苷酸对血管紧张素Ⅱ促VSMC增殖的影响

摘  要:目的探讨脂质体介导的反义表皮生长因子受体寡核苷酸基因转染对血管紧张素Ⅱ促大鼠血管平滑肌细胞增殖的影响.方法用反义表皮生长因子受体寡核苷酸脂质体复合物转染Sprague-Daw ley大鼠血管平滑肌细胞,通过RT-PCR、W estern-B loting分别检测转染后EGFRmRNA及蛋白的表达情况,用3H-Tdr掺入率实验检测经反义表皮生长因子受体寡核苷酸转染再用血管紧张素Ⅱ刺激的血管平滑肌细胞的增殖情况.结果反义表皮生长因子受体寡核苷酸转染大鼠血管平滑肌细胞后,EGFRmRNA及蛋白的表达较正义组及对照组明显减少(P<0.05);用血管紧张素Ⅱ刺激后,反义组细胞的3H-Tdr掺入较正义组及对照组明显降低,经方差分析两者有显著差异(P<0.05).结论脂质体介导的反义表皮生长因子受体寡核苷酸转染减弱了血管紧张素Ⅱ的促血管平滑肌细胞增殖效应,证明了表皮生长因子受体在血管紧张素Ⅱ促血管平滑肌细胞增殖中起重要作用.Objective To investigate the effect of antisense senquences of epidermal growth factor receptor (EGFR) on proliferation of vascular smooth muscle cells (VSMCs) activated by angiotensin Ⅱ (Ang Ⅱ ) . Methods Sprague - Rat VSMCs were transfected with antisense senquences of EGFR. EGFR mRNA and protein in VSMCs were detected by reverse transcription polymerase chain reaction ( RT - PCR) and Western - Bloting. The three groups were stimulated by Ang Ⅱ ; Proliferation of VSMCs was detected by ^3H - Tdr incorperation. Result The expression of EGFR mRNA and protein in VSMCs transfected with antisense senquences of EGFR were signiticantly lower than that transfected with sense senquences of EGFR and control ( P 〈 0.05). The ^3H -Tdr incorperation of VSMCs transfected with antisense senquences of EGFR was markedly lower than that transfected with sense senquences of EGFR and control ( P 〈0. 05) . Conclusion Antisense senquences of EGFR attenuated the proliferation of VSMCs activated by Ang Ⅱ, These indicate that EGFR play an important role in the proliferation of VSMCs activated by Ang Ⅱ.
