摘 要:目的:观察补肾方辅助骨髓干细胞移植治疗mdx小鼠的效果。方法:体外培养正常C57BL/6小鼠骨髓细胞对放疗后mdx小鼠尾静脉移植,用补肾方水提液灌胃实验鼠,观察实验小鼠存活率和移植物抗宿主病(GVHD)症状;移植后4个月/8个月取小鼠骨骼肌以HE染色观察病理状况,以免疫荧光方法检测细胞骨架蛋白(DYS蛋白)表达。结果:补肾方能不同程度降低mdx鼠核中心移位比例和提高DYS蛋白表达量;同时能明显减轻GVHD症状。结论:补肾方能提高骨髓干细胞移植治疗mdx小鼠的效果。Objective: To investigate the effect on Bone marrow stem cell transplantition (BMST) combinated with Bushen fang therapy in mdx mice. Methods: Cultured the bone marrow cells of C56BL/6 in vitro and tranplante these cells to mdx mice after irradiation. Bushen fang was used to cure the mdx mice after BMST. The survivel rate, symptom of Graft Versus Host Disease (GVHD) and motor function of model mice following irradiation and transplantation were observed 4 or 8 months after BMST, prepared the freezesplice of muscles of each group, and made HE staining. The dystrophin (DYS) was detected by SABC immunofluorescene. Results: Bushen fang could decrease the rate of centrally nucleated fibers (CNF) , enhance expressional rate of DYS in different level and relieve the symptom of GVHD in transplanted mice. Conclusion: Bushen fang can improve the therapeutic efficacy of BMST.
