摘 要:通过实验确定了紫外线最适诱变剂量为3min、亚硝基胍的最适诱变剂量150μg/mL以及筛选剂二苯胺、β-紫罗酮、硫酸铜、2-脱氧-D-葡萄糖、氯化锂的筛选剂量分别为0.01g/L、0.5g/L、0.6g/L、1g/L、3g/L。进行了紫外线-氯化锂复合诱变法夫酵母,经发酵培养,获得了一株遗传性状稳定在25℃条件下,虾青素高产的菌株UV-40。Mutagenesis of Phaffia rhodozyma was attempted with UV and NTG. Adaptive dosage of UV and NTG are 3 min and 150 μg/mL. Astaxthin synthesis inhibitors, diphenylamine and β-ionone were used in the selection of astaxthin-overproducing mutants of Phaffia rhodozyma, 3 yeast growth inhibitors, CuSO4, 2-deoxy-D- glucose, LiC1 were tested and their adaptive dosage are 0.01 g/L, 0.5 g/L, 0.6 g/L, 1 g/L, 3 g/L. Strain UV-40 was screened with UV combining LiCl. The result showed that strain UV-40 was good stability and higher capacity of astaxthin under 25 ℃ incubation.
