有机溶剂对黄粉虫N-乙酰-β-D-氨基葡萄糖苷酶活力的影响

摘  要:以黄粉虫为材料,以N-乙酰-β-D-氨基葡萄糖苷酶(NAGase,EC 3.2.1.52)为研究对象.考察了几种有机溶剂对NAGase活力的影响.结果表明:甲醇、乙醇、正丙醇、乙二醇和丙三醇对NAGase的作用均表现为低浓度时对酶有激活作用,随着浓度的升高,激活作用减弱并速转为抑制作用;N,N—二甲基甲酰胺(DMF)和二氧六环对酶的抑制作用呈现明显的浓度效应,随浓度增大酶活力迅速下降.进一步研究二氧六环的抑制作用动力学,结果表明:二氧六环对酶的抑制作用是可逆过程,抑制类型都表现为混合型抑制.测得二氧六环对游离酶的抑制常数K I为8.1%,对酶-底物络合物的抑制常数K IS为30.0%.N - acetyl - β - D - glucosaminidase ( NAGase, EC 3. 2. 1.30) from tenebrio molitor linneeus is ob- tained by extraction with 0. 05 mol/L pH7. 5Tris - HCI and ammonium sulfate fractionation. Effects of different organic solvents on NAGase from tenebrio molitor linneeus is investigated. The results indicate that' the enzyme is activated in methanol, ethanol, propanol, glycol and glycerol solutions at low concentration, but inhibited at high concentration. The effects of DMF and dioxane on the enzyme activity are concentration -dependent. As the concentrations of DMF and dioxane increased, the residual enzyme activity rapidly decreases. The inhibitory kinetic of dioxane on the enzyme is further studis. The result shows that the inhibition reaction of the enzyme in dioxane solutions belongs to reversible process and the inhibition type is determined by Lineweaver -Burk plots to be mixed - type. The inhibition constants of dioxane for NAGase KI and KIS are determined to be 8. 1% and 30. 0%.
