摘 要:本文采用薄层色谱法分析多糖的单糖组成。从斑点分离度、清晰度、有无拖尾现象及展层时间等方面综合评价展层效果。通过实验确定了适合的展开剂为展开系统(乙酸乙脂:吡啶:无水乙醇:水=8:1:1:2)、合适的显色剂为苯胺-二苯胺磷酸显色剂。通过此体系确定了单糖的Rf值,可用于快速分析水解多糖的单糖组分。Applying TLC to analyze monosaccharide composition of polysaccharides was stuided. A mixed solution composed of ethylacetate, pyridine, alcohol and water in a ratio of VC4H8O2o,:VC3H5N:VC2H6:VH8O = 8:1:1:2 by volume is used as extending system. A solution containing aniline and diphenylamine dissolved in acetone is used as coloration system. Satisfactory separation is acquired as shown by the thin layer chromalogram. Qualitative results can be obtained in accordance with the Rf value of the standard spots. Distinct figures indicating the monosaccharide composition of polysaccharides obtained by the modified TLC.The extending system and coloration system can be used in monosaccharide composition analysis.
