尼莫地平对慢性脑缺血大鼠认知和海马CA1区NOS亚型的影响

摘　　要：目的探讨尼莫地平对慢性脑缺血大鼠海马CA1区三种一氧化氮合酶亚型(nNOS、iNOS、eNOS)的表达和认知的影响。方法24只大鼠随机分为假手术组、模型组和治疗组,双侧颈总动脉永久结扎造模,治疗组于术后24h开始用尼莫地平(10mg/kg,2次/d)灌胃,连续60d,各组于60d后用Y型迷宫测试认知功能,测试结束后做免疫组化染色,观察计算海马CA1区每个高倍镜视野下的阳性细胞均数。结果与假手术组相比,模型组认知能力下降,海马CA1区nNOS和eNOS表达下降,iNOS表达增强,治疗组的各项指标介于两者之间。结论尼莫地平能改善慢性脑缺血大鼠的认知能力,其机制可能与提高海马CA1区nNOS和eNOS的表达及抑制iNOS的表达有关。Objective To study the effects of nimodipine on cognition and expression of the three NOS isoforms（nNOS,iNOS, eNOS） of hippocampups CA1 region in rats with chronic cerebral ischemia. Methods 24 rats were randomly divided into three groups. Permanent oeeusion of bilateral common carotid arteries were adopted to made chronic cerebral ischemia rats model. The mimodipine group administered mimodipine（10mg/kg ig. bid） from 24h after operation to 60 days. Then rats of each group were tested the cognitive ability by Y maze. Immunohistochemistry staining was done after the maze test and the mean numbers of positive cells of uNOS,iNOS,eNOS in the CA1 regions of hippocampups were calculated. Results The cognition ability of model group was descending and expression of nNOS and eNOS was less and iNOS was more than the sham operation group. The index of the mimodipine group was more than the latter but less than the former, Conclusion ischemic rats,maybe one machanism is to inhance the expression of region. Nimodipine can improve the cognition ability of chronic cerebral nNOS and eNOS and inhibite that of iNOS in hippocampups CA1
