摘  要:分离得到1株抑制烟草青枯病菌的内生拮抗菌B-001菌株.为确定其抗菌蛋白产生的最佳培养基和培养条件,用硫酸铵沉淀法从发酵液中提取抗菌蛋白质,测定其对烟草青枯病菌抗菌活性,确定产生抗菌蛋白的最佳培养基为BPY液体培养基,最佳发酵条件为pH7.0,30℃,装液量100 mL,200 r/min振荡培养96 h;抗菌蛋白在pH5.0,pH8.0时抑菌活性仍分别保持90%,96%;pH6.0,pH7.0时抑菌活性仍保持不变;对酸、碱稳定;经40~100℃处理20 min后抑菌活性保持不变,具有良好的热稳定性;对蛋白酶有一定的耐受性.在温室内,抗菌蛋白粗提液对烟草青枯病可取得83.8%的防效.A strain of endophytic inhibiting B-001 was isolated, which possesses antagonistic abilities to tobacco Ralstonia solanacearum. In order to make certain what were the optimal medium and fermentative condition in which it made the antagonistic protein, its antagonistic protein was purified by ammonium sulphate precipitation from the fermentative optimal medium, then mensurated antagonistic abilities to tobacco Ralstonia solanacearum. The optimal medium of B-001 is BPY medium, its optimal fermentative condition is pH7,0, 30 ℃, 100 mL/250 mL, 200 r/min, 96 h shaking culture and the antagonistic protein was insensitive to acidic, alkaline such as: at pH5.0, pH8,0 antagonistic abilities were 90%, 96%, at pH6.0, pH7.0 its antagonistic abilities were the same. The antagonistic protein was insensitive to environment heat such as: its antagonistic abilities were the same when it was keeped at 40 - 100 ℃ 20 min, and proteinases under normal temperature.Control efficacy of the crude protein to tobacco bacterial wilt in greenhouse was 83.8% by injection.
