复方奥硝唑甲磺酸培氟沙星缓释牙栓体外释放度的测定

摘　　要：目的测定复方奥硝唑甲磺酸培氟沙星缓释牙栓中奥硝唑、甲磺酸培氟沙星的体外释放动力学。方法采用高效液相色谱法，采用C18色谱柱分离，以甲醇一磷酸二氢钾为流动相，流速1．0ml／min，测定波长277nm，柱温30℃，测定复方奥硝唑甲磺酸培氟沙星缓释牙栓中奥硝唑、甲磺酸培氟沙星的持续释放时间和有效释放浓度。结果①奥硝唑的浓度与峰面积0．00101—0．022mg／ml范围内线性关系良好（r=0．9999），平均回收率为100、09％（rg=5），RSD=0．59％；甲碘酸培氟沙星在0．0053—0．0265rag／ml范围内线性关系良好（r=0．9999），平均回收率为98．84％（rg=5），RSD=2．25％。②13h奥硝唑累和甲磺酸培氟沙星累计释放度分别为98．3％、96．01％。结论复方奥硝唑甲磺酸培氟沙星缓释牙栓具有较长时间的缓释效果。Objective To Determine the dynamic release of omidazole and pefloxacin from the sustained-release periodontal suppository of omidazole and pefloacin by high-performance liquid chromatography （HPLC）in vitro. Methods HPLC analysis was conducted. The operating condition were C18-ODS-A column, methanol-potassium dihydrogen phosphate as mobile phase at a flow rate of 1.0ml/min and a detection wavelength at 277um and a column temperature at 30℃. To detect persistent releasing-time and effective releasing-concentration of ornidazole and pefloxacin from the sustained-release periodontal suppository of ornidazole and pefloxacin. Results ①There was good liner relationship between the concentrations of ornidazole and their peak areas in the ranges of 0.00101 - 0.022 mg/ml（ r = 0.9999） ,The average recovery was 100.09% （ n = 5 RSD = 0.59% ） ;There was good liner relationship between the concentrations of pefloacin and their peak areas in the ranges of 0.053 - 0.0265mg/ml（ r = 0.9999） ,The average recovery was 98.84% （ n = 5 RSD = 2.25 % ）. ②The cumulative release of ornidazole was 98.3 % in 13 hours, and the cumulative release of pefloxacin was 96.01% in 13 hours. Conclusion The sustained-release periodontal suppository of omidazole and pefloxacin have sustained-release effect of long time.
