摘 要:目的:探讨清肝化瘀方药阻断大鼠肝癌前病变的作用机制。方法:SD大鼠随机分为4组:模型组(A组)、清肝化瘀方药等效剂量组(B组)、清肝化瘀方药大剂量组(C组)、对照组(D组),分别用酶组化及免疫组化方法检测γ-谷胺酰转肽酶(γ-GT酶)、Notch1、Notch1配体Delta、NF-κB的表达变化。结果:与D组比较,A组γ-GT酶、Notch1、Delta、NF-κB表达水平显著升高(P<0.01),并且NF-κB与Notch1、Delta之间呈显著正相关(r值分别为0.865、0.742,P<0.01)。与A组比较,C组γ-GT酶、Notch1、Delta、NF-κB表达水平显著下调(P<0.01)。结论:清肝化瘀方药可保护肝细胞阻断癌变,其机制可能与下调Notch1、Delta、NF-κB有关。AIM: To investigate the mechanism of Qinggan Huayu Formula intervening rat's hepatic precancerous lesion. METHODS: SD rats were randomly divided into 4 groups: model group (group A), moderate-dose Qinggan Huayu Formula group (group B ), high-dose Qinggan Huayu Formula (group C ), normal control group (group D), and to detect the expression of γ-GT, Notchl ,Delta, NF-kB by enzyme-histochemisty and immunohistochemistry methods. RESULTS : Compared with group D , the expressions of γ-GT, Notchl, Delta, NF-kB in group A were singnificanfly increased ( P 〈 0.01 ), There were singnificantly positive correlations among NF-kB, Notchl and Deha(P 〈0.01 ) ,Compared with group A expressions of γ-GT,Notchl ,Delta, NF-kB in group C was singnificantly increased(P 〈0.01 ). CONCLUSION: Qinggan Huayu Formula can protect from hepatic precancerous lesion,the mechanism was related to regulate the expressions of Notch1 ,Delta and NF-kB.
