摘  要:目的研究钙离子非依赖型磷酸酯酶A2,IVA亚型(iPLA2β)在视网膜色素上皮细胞(Retina pigment epitheli-um,RPE)吞噬光感受器脱落膜盘(photoreceptor outer segments,POS)过程中所起到的作用,继而探讨其对老年性黄斑病变(Age-related macular degeneration,AMD)的发病过程影响。方法应用western Blotting方法检测ARPE-19细胞吞噬牛POS后iPLA2β表达含量的变化。利用RNA干扰技术,即iPLA2β干扰RNA的质粒构建的ARPE-19细胞进行吞噬功能检测,从而评价iPLA2β在RPE细胞吞噬过程中所起的作用。结果我们发现ARPE-19细胞吞噬POS后,iPLA2β的蛋白表达增强。经iPLA2βSiRNA转染的ARPE-19细胞吞噬功能明显降低。结论iPLA2β对于RPE细胞的吞噬作用起着重要的调节作用,它可能在光感受器细胞的更新和良好视力的维持过程中起到重要的作用。RPE细胞的功能缺陷又是AMD的主要发病原因,对于iPLA2β的功能和它在RPE细胞吞噬作用中所起到的作用的理解可以对此类疾病的治疗提供非常有价值的线索。Objective The aim of this project was to explore the involvement of iPLA2β in the photoreceptor outer segment (POS) phagocytosis by human retinal pigment epithelial cells (RPE), discuss the possible role of iPLA2β in the pathogenesis of AMD.Methods Westem blotting was used to observe the expression of iPLA2β induced by the phagocytosis of bovine POS by ARPE-19 cell line.Small interference RNA method with iPLA2β SiRNA was applied to evaluate the role of iPLA2β in RPE Phagocytosis.Results Our studies showed iPLA2β expression was upregulated after the phagocytosis of the POS by RPE cells.Furthermore,the phagoeytosis was decreased after transfection with iPLA2β SiRNA vector. Conclusion iPLA2β is very important for the regulation of the Phagocytosis of POS by RPE cells and it might play a critical role in the pathogenesis of AMD, which is a serious ocular disease caused by RPE dysfunction.
