摘 要:以黑曲霉基因组DNA、mRNA为模板,利用种属相似性设计了一对引物,采用PCR与RT—PCR技术得到伊葡萄糖苷酶基因的DNA和cDNA全序列.测序表明:克隆得到的DNA序列全长2924bp,cDNA序列全长2583bp,编码860个氨基酸.编码序列推导的蛋白质序列相似性分析显示:完整蛋白质序列同源性高达97%~99%.该研究为β-葡萄糖苷酶基因的遗传转化及应用奠定了基础。Genomic DNA and mRNA were isolated from Aspergillus niger, and the high efficient primers was designed by species similarity. With the methods of PCR and RTPCR respectively, the fragments of the DNA and cDNA of beta-glucosidase gene were obtained. Seqence analysis indicated that DNA fragment was 2 924 bp, and the cDNA sequence was 2 583 bp, which encode 860 amino acids. The identity of the derived protein with Aspergillus niger ffglucosidase has the high homogeneous characteristics of 97%-99%. The foundmental researches for transformation and application of this gene have been achieved in this paper.
