摘　　要：目的探讨cyclinD1/D2/D3在大鼠脑胶质瘤组织中的表达。方法将体外培养的大鼠C6胶质瘤细胞(细胞数为3×105个)借助动物立体定向仪接种于Wistar大鼠左侧尾状核区,解剖标本,行组织病理学检查及GFAP、cyclinD1/D2/D3免疫组化检测。结果cyclinD1/D2/D3均呈阳性表达。cyclinD1/D3主要在细胞胞核表达,cyclinD2主要在胞浆表达,部分在胞核表达,不同存活期荷瘤鼠瘤组织中cyclinD1/D2/D3表达量不同。结论cyclinD1/D2/D3过分表达参与调节大鼠脑胶质瘤增殖。荷瘤鼠生存期与cyclinD家族蛋白的表达呈负相关关系。Objective This experiment was designed to detect the expression of cyclin D1/D2/D3 in the tumor tissues of rat glioma models. Methods The C6 cells cultured in vitro were stereotaxically implanted into the left caudate nucleus of Wistar. The concentration of C6 cells was 5 × 10^9/L （the injecting volume was 60 μl for each rat ）. After C6 inoculation, the living and survival of animals were observed. Tumor was confirmed with HE and immunohistochemistry staining of GFAP. Cyclin D1/D2/D3 were detected by immunohistochemistry. Results The study demonstrated the expression of cyclin D in glioma tissues. Immunohistochemical reaction of cyclin D1/D3 was confined to the nucleus, while the expression of cyclin D2 was mainly in cytoplasm. There was slight difference of expression of all kinds of cyclin D among three study groups. Conclusion The data suggest that amplification and increased expression of cyclin D family contribute to the loss of cell cycle control in a small fraction of Wistar C6 gliomas. The overexpression of cyclin D family is negatively related with survival time of the rats with tumors.
