摘  要:目的观察即早基因c-jun mRNA在多发性梗死性痴呆大鼠发病不同时间点的表达,探讨c-jun基因在多发性梗死性痴呆发生发展中的病理生理机制。方法采用改良的微栓子法(取自体血栓注入颈内动脉)制作大鼠多发性梗死性痴呆模型。应用原位杂交法检测大鼠多发性梗死性痴呆发病不同时间点c-jun mRNA的表达情况。结果正常大鼠大脑皮层和海马区内即有少量的c-jun mRNA的表达,假手术组的表达略高于正常组(P>0.05)。多发性梗死性痴呆大鼠c-jun mRNA阳性细胞平均光密度值明显高于假手术组及正常对照组(P<0.05)。动态观察发现,c-jun mRNA在大鼠多发性脑梗死后2 h开始高表达,4 h达高峰,1W后开始下降。结论多发性梗死性痴呆即早基因c-jun mRNA呈规律性高表达。Objective To observe the expression of c-jun mRNA in multiple infarct dementia (MID) rats at different time-points to explore the pathophysiologieal mechanism of MID. Methods Omogenous mierothrombi were injected into internal carotid artery to make cerebral multi-infarction dementia model. The expression of c-jun mRNA was determined by in-situ hybridization at different time points after infarction. Results c-jun mRNA expressed a little in cortex and hippocampus of normal rats, and increased a little in sham group than in normal rats( P 〉 0.05). The average optic density(AOD) of c-jun mRNA positive cells increased greatly in MID with significant difference compared to the normal control group and the sham group( P 〈 0.05 ). The AOD value of c-jun mRNA positive cells increased 2 hrs post-infarction, to the peak 4 hrs after infarction, kept on 1 week( P 〈 0.05). Conclusion c-jun mRNA showed a regular overexpreasion in cortex and hippocampus of MID rats.
