摘 要:目的探讨表皮生长因子受体在血管紧张素Ⅱ促大鼠血管平滑肌细胞增殖效应中的作用。方法用反义表皮生长因子受体寡核苷酸脂质体复合物转染SD大鼠血管平滑肌细胞,用逆转录聚合酶链反应、Western-Blot-ing分别检测转染后表皮生长因子受体mRNA及蛋白的表达情况,用氚标胸腺嘧啶脱氧核苷掺入实验检测血管平滑肌细胞的增殖情况。结果反义组大鼠血管平滑肌细胞表皮生长因子受体mRNA表达(0.18±0.03)较正义组(0.61±0.11)及对照组(0.66±0.09)明显减少(P<0.05),反义组大鼠血管平滑肌细胞表皮生长因子受体蛋白的表达(43.1±8.4)较正义组(92.6±10.5)及对照组(100.7±11.3)明显减少(P<0.05);反义组细胞的氚标胸腺嘧啶脱氧核苷掺入率(1055.1±95.7)较正义组(1882.4±129.7)及对照组(2013.3±121.3)明显降低(P<0.05)。结论表皮生长因子受体在血管紧张素Ⅱ促血管平滑肌细胞增殖中起重要作用。Aim To investigate the role of epidermal growth factor receptor (EGFR) in proliferation of vascular smooth muscle cells (VSMC) activated by angiotensin Ⅱ (AngⅡ). Methods The cultured VSMC of Sprague-Dawley rat were transfered with antisense sequences of EGFR. EGFR mRNA and protein in VSMC was detected by reverse transcription polymerase chain reaction (RT-PCR) and western blot analysis, the proliferation of VSMC was determined by 3H-deoxythymidine (3H-TdR) incorporation. Results The expression of EGFR mRNA in VSMC transfected with antisense sequences of ECFR were significantly lower than transfected with sense sequances of EGFR and control group (0.18±0.03, 0.61±0. 11, and 0.66±0.09 respectively, P〈0.05).The expleasion of EGFR protein in VSMC tramfected with anfisense sequences of EGFR were significantly knver than transfected with seine sequances of EGFR and control group (43.1 ±8.4, 92.6± 10.5, and 100.7± 11.3 respectively, P〈0.05).The H-TdR incorporation of VSMC transfered with antisense sequences of EGFR were significantly lower than that transfered with sense sequances of ECFR and control group (1 055.1±95.7, 1882.4±129.7, and 2013.3±121.3 respectively, P〈0.05).Conclusion EGFR play an important role in proliferation d VSMC d rat induced by anglotensin Ⅱ.
