摘 要:目的研究心肌营养素1在腹主动脉缩窄所致大鼠压力负荷性心肌肥大模型中的作用,观察AG490对心肌肥大及心肌营养素1表达的影响。方法用腹主动脉缩窄的方法建立心肌肥大大鼠模型,检测左心室重量指数,应用逆转录聚合酶链反应检测心肌营养素1、β-肌球蛋白重链mRNA表达,放射免疫分析法测定血浆和心肌血管中血管紧张素Ⅱ水平。结果腹主动脉缩窄术后14天心肌肥大组左心室重量指数明显高于假手术组(P<0.01);AG490干预组左心室重量指数明显低于心肌肥大组(P<0.01),但仍高于假手术组(P<0.01)。心肌肥大组血浆和心肌血管紧张素Ⅱ增高,与假手术组比差异有统计学意义(P<0.01);AG490干预组血管紧张素Ⅱ水平低于心肌肥大组(P<0.05),但仍高于假手术组(P<0.01)。心肌肥大组心肌营养素1mRNA表达较假手术组增加(P<0.01);AG490干预组心肌营养素1表达较心肌肥大组无明显变化(P>0.05),但与假手术组相比表达明显增加(P<0.01)。心肌肥大组心肌β-肌球蛋白重链mRNA表达明显高于假手术组(P<0.01);AG490干预组β-肌球蛋白重链mRNA表达较心肌肥大组明显降低(P<0.01),与假手术组相比表达仍增加(P<0.01)。结论心肌营养素1参与腹主动脉缩窄所致大鼠压力负荷性心肌肥大的发病过程,AG490可抑制心肌营养素1的表达及心肌肥大。Aim To investigate the role of cardiotrophin-1 (CT-1) in the rat model of pressure overload-induced cardiac hypertrophy and the effect of AG490 on the cardiac hypertrophy and CT-1. Methods The model of pressure overload was established by constriction of abdominal aorta. The left ventricular mass index (LVMI) was observed. The mRNA levels of CT-1 and β-myosin heavy chain (β-MHC) were examined by reverse transcription polymerase chain reaction (RT-PCR) method. The circulating and ventricular angiotensinⅡ (Ang Ⅱ) was measured by radiommoassay. Results Fourteen days after constriction of abdominal aorta, the LVMI of the hypertrophic group was significantly increased than that of the control group ( P 〈 0.01 ) and it was significantly decreased in AG490 group than in the hypertrophic group, but more than in the control group ( P 〈 0.01 ). The circulating and ventricular Angll of the hypertrophic group was increased compared with the control group, and there was a significant difference between the two groups ( P 〈 0.01 ) ; the Angll level of AG490 group was less than that of the hypertrophic group, but still more than the control group ( P 〈 0.01 ). The expreasion of CT-1 mRNA was similar in the hypertrophic group and the AG490 group (P〉0.05), and beth groups were more than the control group (P〈0.01). The expreasion of β-MHC mRNA in the hypertrophic group was significantly increased than that in the control group ( P 〈0.01 ) and decreased in the AG490 group compared with the hypertrophic group ( P 〈0.01), but it was still move than the control group ( P 〈 0.01) sions The expression of CT-1 may play an important role in cardiac hypertrophy, AG490 may inhibit the expression of CT-1 and the development of cardiac hypertrophy.
