一株产琼胶酶土壤细菌Paenibacillus sp. MY03的筛选与分子鉴定

摘　　要：为寻找高产琼胶酶的微生物新资源,促进琼胶低聚糖的研究与开发,本研究自多个土壤样品中进行微生物的分离,借助于卢戈氏碘液染色法进行琼胶酶生产菌的筛选,通过DNS-还原糖测定比较不同微生物的产酶特性,克隆、分析细菌的16S rRNA基因序列进行分子鉴定。最后得到1株胞外琼胶酶活性较高的土壤细菌,鉴定至Paenibacillus属,命名为MY03菌株;分子鉴定表明Paenibacillus sp. MY03是产琼胶酶细菌家族中的新成员。In order to promote the research and exploitation of agar - derived oligosaeeharides, soil microbe were isolated and screened for new agarase - producing bacterial resources. By employing the 16S rRNA gene method, an isolate named MY03 has been identified to be a member of the Paenibaeillus group. The Paenibaeillus sp. MY03 was also a new member of the agarase - producing bacterial family, according to the molecular characterization in this study.
