摘  要:目的:了解回流消化技术在食品蛋白质含量测定中的应用效果。方法:采用自动回流消化仪替代国标法蛋白质测定中使用凯氏定氮烧瓶进行消化的方法。由于样品与试剂的反应是在相对密闭的容器内进行,所以在消化过程中分解产生的大量酸雾、硫的氧化物等刺激性气体溶于冷凝水中排出,对环境的污染和对分析人员毒害大大减小。结果:实验结果表明,回流消化法与国标法比较,测定结果基本一致,t检验无显著性差异(P>0.05),测定样品蛋白质含量在1.59%~23.55%之间的相对标准偏差(RSD)为0.64%~1.96%(n=5),平均回收率为98.3%。结论:自动回流消化法仪器价格便宜,不需要消毒通风柜,可节约经费开支,消化时间较为缩短,减轻了分析人员的劳动强度,相对提高了工作效率。对基层卫生检验实验室具有一定的推广应用价值。Objective: To determine the value of reflux digestion for determination of protein content in food. Methods: The automatic reflux digestion device was used to replace the reagent used in International Method. Since the reaction of samples with reagents existed in a relatively close container,the acid fog and oxides of sulphur produced by the digestion were resolved in the congealed water to be discharged. Consequently, their harm to environment was greatly reduced. Results:The determined results by reflux digestion were in consistent with those by the International Method ( P 〉 0. 05 ). The determination RSD was 1.59% ~ 23. 55% and the average recovery was 0. 64% ~ 1.96%. Conclusion :The automatic reflux digestion is a method of low cost and high efficiency. Therefore, it can be used widely in low - level health loboratories.
