摘 要:目的:观察反义寡核苷酸对损伤视神经Nogo-A的影响。方法:实验分为对照组、随机序列组、和2,5,10μmol/L3种Nogo-A反义寡核苷酸浓度组。大鼠视神经钳夹伤后,采用逆转录-聚合酶链反应法(RT-PCR),半定量分析反义寡核苷酸对损伤视神经Nogo-A mRNA表达量的变化。结果:反义寡核苷酸均显著降低Nogo-A mRNA的表达(P<0.01),随机序列对Nogo-A mRNA的表达无影响(P>0.05)。结论:反义寡核苷酸对视神经损伤后Nogo-A mRNA表达有抑制作用。AIM: To observe the effects of antisense oligodeoxynudeotides (ASODN) on Nogo-A mlRNA expression in injured optic nerves. METHODS: A total of 30 rats were divided into five groups: control group, random sequence group and test groups (2μmol/L, 51μmol/L and 101μmol/L Nogo-A antisense oligodeoxynudeotides), reverse transcriptase-polymerase chain reaction (RT-PCR) method was used in the detection of the levels of Nogo-A mRNA in injured optic nerves. RESULTS: The result of RT-PCR showed that Nogo-A antisense oligodeoxynudeotides could significantly inhibit the expresson of Nogo-A mRNA (P〈0.01), random sequence antisense oligedeoxynudeotides had no noticeable effect on Nogo-A mRNA. CONCLUSION: Nogo-A antisense oligodeoxynudeotides can inhibit the expresson of Nogo-A mRNA in injured opticnerves.
