牛磺酸对糖尿病大鼠视网膜Mller细胞凋亡的防护研究

摘　　要：目的观察高糖培养条件及牛磺酸干预视网膜Mller细胞的凋亡变化,探讨牛磺酸对早期糖尿病视网膜Mller细胞的保护作用。方法取出生后3d的SD大鼠视网膜,分离纯化培养视网膜Mller细胞;经预实验选取25mmol/L葡萄糖作为高糖培养条件,实验分为6组:正常组;高糖+0.1mmol/L牛磺酸组;高糖+1mmol/L牛磺酸组;高糖+5mmol/L牛磺酸组;高糖+10mmol/L牛磺酸组,用原位缺口末端标记法(TUNEL)法检测视网膜Mller细胞凋亡并观察牛磺酸的防护效应。结果神经胶质酸性蛋白(glia1fibrillary acid protein,GFAP)和S-100荧光双染鉴定纯化培养的视网膜Mller细胞均被双染;牛磺酸干预后TUNEL阳性细胞数量明显减少,并呈一定剂量的关系效应,1～10mmol/L牛磺酸干预TUNEL阳性细胞显著低于高糖培养组(P<0·01)。结论牛磺酸能够呈剂量依赖性地抑制高糖所致视网膜Mller细胞凋亡。Objective To observe the apoptosis changes of retinal MUller cells clutured in high glucose after treated with taufine and further investigate the protection of taufine on Mueller cells in early diabetic retinopathy. Methods MUller cells were separated and purified from the retinas of 3-day SD rats and cultured in 25 mmol/L glucose that was proved to be high glucose culture for MUller cells by pre-expefiment, and according to the taurine dose for treatment divided into 5 groups： 0, 0.1, 1, 5, 10 mmol/L taurine. The MUller cells not cultured in 25 mmol/L glucose and without taurine treatment served as normal control. The apoptosis of retinal Mttller cells was detected by TUNEL and the protective effects of taurine on Mueller cells in high glucose was observed. Results The MUller cells were identified by double staining with Glial Fibrillary Acid Protein and S- 100 in immunocytochemical fluorescein test. The positive cells decreased significantly after the taurine treatment, in a dose dependent manner. The amount of positive cells in taurine-treated group （ 1,5, 10 mmol/L taufine） was less than that of high glucose group （0 mmol/L taurine） （P 〈0. 01 ）. Conclusion Taurine can inhibit the apeptosis of retinal Muller cells induced by high glucose and the inhibition effect is related to taurine dose.
