摘  要:Objective To investigate the change of high mobility group box-1 protein (HMGB1) level in the liver and the effect of Xuebijing injection on hepatic tissues high mobility group box-1 protein (HMGB1) expression and acute liver injury in rats with delayed resuscitation after scald injury. Methods Wistar rats were subjected to 30% full-thickness scald injury followed with delayed resuscitation. Hepatic tissues samples and blood samples were harvested to determine HMGB 1 mRNA as well as protein expression and organ function parameters. HMGBlmRNA level was semi-quantitatively measured by the reverse transcription polymerase chain reaction (RT-PCR) taking GAPDH as an internal standard, and protein expression of HMGB 1 was detected by Western blot. The heoatic tissues functional indices--serum ALT and AST contents, were measured with automatic biochemistry analyzer. Results Compared with sham scald controls, both mRNA and protein expressions of HMGB1 were significantly enhanced in the hepatic at 8-72h after scald injury (P〈0.05 or P〈0.01), meanwhile level of ALT and AST of serum were markedly increased after scald (P〈0.05 or P〈0.01).Treatment with Xuebijing could markedly down-regulated hepatic HMGB1 mRNA and protein expressions at 24h, 48h and 72h (P〈0.05 or P〈0.01), and reduced serum level of ALT and AST following scald (P〈0.05). Conclusion These data indicated that HMGB 1 appears to be involved in the pathogenesis of post scald acute hepatic injury. Xuebijing administered inhibits hepatic tissues HMGB 1 expression, and protects hepatic against acute injury induced by scald injury.
