摘 要:建立了鹿茸超临界CO2萃取物中胆固醇含量的测定方法。采用气相色谱法测定,FID检测器;柱程序升温:278℃保持5min,以2.5℃/min升至286℃,保持6min,后以40℃/min升至305℃并保持3min;气化室温度:295℃;检测器温度:305℃;氮气流速:30ml/min;氢气流速:47ml/min;空气流速:400ml/min。结果表明,胆固醇在0.20~1.00μg与吸光度呈良好的线性关系,回归方程为Y=106X-54842(r=0.9989),加样平均回收率为100.4%,RSD为3.3%(n=5)。A method for determination of cholesterol in extracts from antler velvet by supercritical carbon dioxide (SC-CO2) extraction was presented. Cholesterol was assayed by gas chromatography with flame ionization detection, where oven temperature was controlled with a temperature elevation program set at 278 ℃ for 5 min, elevated to 286 ℃ at the rate of 2.5 ℃/min for 6 min, and then elevated to 305 ℃ at the rate of 40 ℃/min maintained for 3 rain. The temperature at injector port and detector was set at 295 ℃ and 305 ℃ respectively. The flow rates were 30 ml/min for nitrogen, 47 ml/min for hydrogen, and 400 ml/min for air. The calibration curve is linear with the ranges of 0.20- 1.00 ×g for cholesterol and the regression equation is Y=10^6X- 54842(r=-0.9989). The recovery is 100.4% with RSD 3.3%(n=5).
