摘  要:目的比较几种氨基酸溶液对卵巢癌多细胞球体(MCS)生长的影响。方法分别用含有平衡氨基酸、增量精氨酸、去蛋氨酸、去缬氨酸、复合氨基酸(增量精氨酸、去蛋氨酸、去缬氨酸)营养液的培养基培养卵巢癌MCS,观察并分析生长曲线、细胞周期,检测增殖细胞核抗原(PCNA)的表达。结果生长曲线可见平衡氨基酸组细胞生长速度明显高于其他各组,复合组细胞生长速度最慢,与其他组比较,差异有统计学意义(P<0.05)。细胞周期可见平衡氨基酸组细胞多数处于S期,处于G0/G1期的细胞含量为(41.5±6.3)%,而复合组处于G0/G1期的为(77.6±4.7)%,与其他各组比较,差异有统计学意义(P<0.05)。PCNA检测结果表明,复合组PCNA的转录水平低于其他各组。结论平衡氨基酸有利于卵巢癌MCS的生长;4种不平衡氨基酸对MCS的生长均有抑制作用,并能阻滞MCS细胞进入增殖期,其中复合氨基酸的作用最强。Objective To study the effect of diffierent amino acid(AA) solutions on growth of multiple cellular spheroid (MCS) of ovarian cancer. Methods Different AA were applied to MCS, including balance AA, arginine- enriched, methionine - depleted, valine - depleted and complex AA which was composed of arginine - enriched, methionine - depleted and valine - depleted. Observe and analyze growth curve and cell cycle, and detect expression of proliferation cell nuclear antigen( PCNA). Results MCS in balance AA grew most fast, while MCS in complex AA grew most slowly, which was significantly different from other groups(P〈0.05). Most of the cells in balance AA were in S phase, the number of cells in G0/G1 phase was (41.5 ± 6.3) %. Most of the cells in complex AA were in G0/G1 phase[(77.6 ± 4.7) %], which was signficantly different from other groups(P 〈 0.05). Expression of PCNA of cells in complex AA was lower than other groups. Conclusion Balance AA can stimulate MCS to grow. The four kinds of imbalance AA can inhibit MCS and block them into proliferative phase, and complex AA has the most significant effect.
