摘  要:目的用染色体8p微卫星的杂合性缺失(LOH)作为肿瘤的遗传性标记,对同一肝细胞癌结节内部不同肿瘤细胞群是否有克隆差异进行研究。方法在10例肝细胞癌患者,同一肿瘤结节内部不同颜色质地的部分作为不同的细胞群,手工辅助显微切割获取较纯的肿瘤细胞,抽提肿瘤细胞DNA;选取5个染色体8p微卫星位点D8S277、D8S254、D8S258、D8S298、D8S1771,进行微卫星LOH的检测。结果在10例肝细胞癌患者,7例检测出8p微卫星的LOH,3例未检测出8p微卫星的LOH。6例患者在同一肿瘤结节的内部不同肿瘤细胞群之间出现微卫星位点LOH类型的差异,1例患者3个肿瘤细胞群的LOH类型相同。结论在同一肝细胞癌结节的内部,不同的肿瘤细胞群之间存在克隆差异,这种差异说明不同的肿瘤细胞群可能有不同的克隆来源,从而为观察同一肿瘤内不同肿瘤细胞群的生物学特性提供依据。Objective The loss of heterozygosity (LOH) of chromosome 8p microsatellite was used as the genetic marker to identify the clonal disparity among different cell groups in the same hepatocellular carcinoma (HCC) nodule. Methods In 10 patients with HCC,the parts with different colors in the same nodule were taken as different cell groups; Relative pure tumor cells were obtained by hand-assistant microdissection; LOH of 5 microsatellites in chromosome 8p was detected. Results LOH was detected in 7 patients, and 6 of them had different LOH alterations among different cell groups in the same tumor nodule. One had 3 different cancer cell groups with same LOH pattern. Conclusion In the same HCC nodule, different cell groups have dissimilar LOH, indicating that there are clonal differences among these cell groups and maybe the different cell groups have different clonal origins.
