摘 要:采用离子交换法吸附发酵液中的丁酸,从18种树脂中筛选出了1种对丁酸吸交性能较好的树脂A。树脂A对10g/L的丁酸溶液中丁酸的静态吸附量为243.3g/kg,对5g/L丁酸梭状芽孢杆菌发酵液中丁酸的静态吸附量则为142.2g/kg。动态吸附实验表明,树脂A吸附性能良好,洗脱峰高而窄,再生性能优良。5L发酵罐实验表明,发酵残液经陶瓷膜过滤,再经过离子交换树脂脱酸后,补充新鲜培养基打入发酵罐中进行循环使用,大大降低了原发酵液中丁酸对丁酸梭状芽孢杆菌生长的抑制,大幅度提高了菌体浓度,使之由补料分批高浓度培养时的6.1×10^8个/mL提高到1.2×10^9个/mL。ABSTRACT High-density culture must be carried out before the preparation of microecological agents. We used ion-exchange method to remove butyric acid from fermentation broth..An appropriate resin A was select- ed from more than 18 types of resins. The static state absorption capacity of Resin A was 243. 3 g/kg from 5g/L butyric acid solution and 142.2g/kg from 6g/L fermentation broth. The dynamic state exchange trials, including adsorption,elute and regeneration of the resin was performed. After removing of butyric acid from fermentation broth using ion-exchange method,we studied high cell density culture with fed-batch culture in bioreactor. Cell concentration of Clostridium butyricurn in5L bioreactor high-density culture increased from 6. 1×10^8 个/mL to 1.2×10^9 个/mL.
