固定化胰蛋白酶壳聚糖树脂的制备及其对大豆胰蛋白酶抑制剂的吸附性

摘  要:通过反向悬浮交联法制备了壳聚糖树脂,采用环氧氯丙烷活化,对树脂固定胰蛋白酶进行研究。通过正交设计实验,优化了壳聚糖树脂的活化、偶联条件;结果表明,4.0g壳聚糖树脂,加入1.6mL环氧氯丙烷,8.0mL1.0mol/LNaOH,活化温度60℃、偶联温度30℃时,胰蛋白酶可达到较大偶联量,偶联量为24.63mg/g壳聚糖树脂;添加硼氢化钠4.0mg,1,4-二氧六环6.0mL时,可将壳聚糖树脂的活化效果分别提高2倍、1倍。固定化酶活性检测表明,胰蛋白酶已成功地偶联到壳聚糖树脂上,并具有亲和吸附胰蛋白酶抑制剂活性,对大豆胰蛋白酶抑制剂的清除率约为33.3%。此固定化胰蛋白酶树脂可作为一种新型、廉价的亲和载体材料。The trypsin immobilized chitosan resin was prepared by epichlorohydrin. The optimum condition on activating and coupling of chitosan resin was obtained by means of orthogonal design experiments. By using the BANA method, the best parameters of technology were found as follows: 4.0g chitosan resin mixed with 1.6mL epichlorohydrin and 8.0mL of 1.0mol/L NaOH, activated at 60℃, coupled at 30℃, the trypsin could get a better coupling capacity, and the coupling capacity was 24.63mg/g chitosan resin. The activation can be improved two times and one times by adding 4.0mg sodium borohydride and 6.0mL 1,4-dioxane respectively. The determination results of immobilization trypsin activity showed that the trypsin had coupled with chitosan resin, and immobilized trypsin chitosan resin had the adsorption affinity to soybean trypsin inhibitor.The scavenging rate for soybean trypsin inhibitor was 33.3%. The immobilization trypsin chitosan can be a new cheap affinity carrier material.
