环氧合酶-2抑制剂诱导宫颈癌Hela细胞的凋亡及相关分子机制的研究

摘　　要：目的:探讨环氧合酶-2(COX-2)抑制剂NS-398对宫颈癌Hela细胞株增殖和凋亡的作用机制。方法:用不同浓度(100-400μmol/L)的NS-398,以不同时间段(12-72h)处理宫颈癌Hela细胞,以四甲基偶氮唑蓝(MTT)法测定Hela细胞增殖抑制率;流式细胞术(FCM)检测Hela细胞凋亡率;Western blot检测COX-2蛋白的表达;放射免疫法检测Hela细胞PGE2释放水平。结果:不同浓度的NS-398(100-400μmol/L)对Hela细胞的增殖具有抑制作用,并呈明显的时效和量效关系,与对照组相比,差异有极显著性(P<0.01)。200-400μmol/L的NS-398诱导Hela细胞的凋亡率,与对照组比较,差异有极显著性(P<0.01),并呈浓度依赖性。NS-398能明显抑制Hela细胞COX-2的活性和表达,随着浓度的增加,其COX-2蛋白表达逐渐降低,与对照组相比,差异有显著性(P<0.05)。不同浓度的NS-398(100-400μmol/L)可明显抑制Hela细胞PGE2释放水平,呈浓度依赖性,与对照组比较,差异有极显著性(P<0.01)。结论:NS-398对宫颈癌Hela细胞生长具有抑制作用,并促进其凋亡,其作用可能与抑制COX-2活性表达及抑制细胞PGE2释放水平有关。Objective： To investigate the Hela cells growth inhibition and apoptosis induced by selective cyclooxygenase-2 （COX-2） inhibitor NS-398. Methods： Hela cells were treated with various concentrations （100- 400μmol/L） of NS-398 at different time. Cell growth was measured by MTT. Apoptosis was detected by double staining flow cytometry （FCM）. Levels of PGE2 were measured by radioimmunoassay. The expression of COX-2 protein was examined by Western blot analysis. Results： After treated with different concentrations of NS-398, the growth of Hela cells was suppressed significantly in a dose-and time-dependent manner （P〈0.01）. The NS-398 can induce apoptosis in a dose-dependent manner. The release of PGE2 was reduced in Hela cells and was significantly different compared with control group （P〈 0.01）. The NS-398 could inhibit the activity and expression of COX-2 in a dose-dependent manner and down-regulated the expression of COX-2 protein greatly. Conclusion： NS-398 could inhibit the proliferation and increase apoptosis in human Hela cells. These effects may be depend on the inhibition of the expression of COX-2 and PGE2 by NS-398.
