摘　　要：目的:研究幽门螺杆菌(Helicobacter pylori,Hp)对胃癌细胞中的RhoA的表达和活性的影响,探讨Hp与胃癌之间的相互关系。方法:Hp由人胃黏膜组织中分离,固体培养,收获后用超声波方法裂解Hp,以裂解液刺激胃癌细胞SGC-7901,用pull-down方法来获得细胞裂解液中的活性RhoA,用Western blot方法来检测细胞裂解蛋白中的活性RhoA的变化,用逆转录聚合酶链反应(RT-PCR)方法半定量法测定RhoA mRNA相对量。结果:用Hp裂解液刺激细胞10 min,总RhoA无明显变化,而活性RhoA升高。结论:Hp裂解液能增加胃癌细胞中活性RhoA的表达,呈剂量-效应关系,但总RhoA无明显变化,它可能是参与胃癌细胞的迁移。　Objective： The experiment was designed to study the effect Helicobacter pylori（Hp） on the activity of RhoA in the gastric cancer cells and investigate the relation between Hp and gastric cancer.Methods： Hp was isolated from human gastric mucosa and cultured on solid culture plate.The lysate of Hp was obtained by sonication.After stimulating the gastric cancer cell line SGC-7901 for ten minutes by the lysate of Hp,we obtained the active RhoA by pull-down assay and examined the level of active RhoA by western blot.The mRNA level of RhoA gene was examined by semi-quantitative RT-PCR. Results： In the gastric cancer cells stimulated by the lysate of Hp for ten minutes,the level of active RhoA was increased but the mRNA level did not change. Conclusion： The lysate of Hp could stimulate the expression of active RhoA dose-dependently,but the expression of RhoA was not increased,it may participate the migration of the gastric cancer cells.
