摘  要:目的:观察表没食子儿茶素没食子酸酯(EGCG)诱导人肝癌(SMMC-7721)细胞系,细胞凋亡的生物学效应。方法:通过MTT比色法检测EGCG对SMMC-7721细胞生长的影响;应用细胞计数法研究不同浓度EGCG对SMMC-7721细胞生长曲线的影响;PI染色流式细胞术研究EGCG对SMMC-7721细胞周期和凋亡率的影响。结果:EGCG显著抑制人肝癌SMMC-7721细胞的生长,呈浓度依赖性;SMMC-7721细胞经EGCG作用后,其细胞群体倍增时间明显延长,细胞增殖周期延长,从而细胞增殖速度减慢;流式细胞术分析结果表明EGCG(30μg/mL、60μg/mL、120μg/mL)处理SMMC-7721细胞48h后,G1期细胞累积均逐渐增加。结论:EGCG具有诱导人肝癌SMMC-7721细胞凋亡的作用,其机制可能与使细胞周期G1期阻滞相关。Objective:To investigate the effect of epigallocatechin - 3 - gallate on induction of apoptosis in liver cancer cell line SMMC - 7721. Methods:The growth inhibition of SMMC - 7721 cells was measured by MTT assay. Cell couting method was used to test the influence of growth curve in SMMC - 7721 cells treated with different concentrations of EGCG. The apoptosis rate and cell cyle of SMMC - 7721 cells were determined by PI staining flow cytometry. Results: EGCG significantly inhibited the growth of SMMC- 7721 cells,in a concentration dependent manner. Cell counting showed that average doubling time increased by EGCG in a concentration dependent manner. PI staining flow cytometry showed that cell cyle was arrested at G1 phase and the apoptosis of SMMC -7721 cells was induced by EGCG in a concentration dependent manner. Conclusion:EGCG posseses antitumor effect in vitro,its mechanism might be associated with G1 phase cell cycle arrest.
