扫描电镜下乙醇对兔角膜上皮细胞活性的影响

摘　　要：目的 观察LASEK术中乙醇对兔角膜上皮细胞活性的影响，指导LASEK术中选择适宜的乙醇浸润时间。方法 10只实验兔随机分为5组，每组2只：正常对照组、20s组、30s组、40s组、50s组，20％乙醇浸润时间为20s、30s、40s、50s。角膜标本行锥蓝染色以及扫描电镜观察。结果 20％乙醇浸润兔角膜上皮细胞20s，细胞活性无明显降低；浸润时间一旦超过30s，细胞活性明显降低。结论 LASEK术中20％乙醇的最适浸润时间为20s左右，不要超过30s。Objective To observe the vitality of corneal epithelieum after exposed to 20% alcoholic solution for different time. Methods According to the time of rabbits sacrifice,10 healthy adult New Zealand white rabbits were devided into 5 groups randomly.They were fresh control group, 20s group,30s group,40s group and 50s group. In experimental groups the eyes were exposed to 20% alcoholic solution for 20s,30s,40s or 50s. Trypan blue staining, scanning electron microscopes were used to study the structure of corneal epithelial cells. Results Contrast with the fresh control group ,the corneal epithelial cells which were infiltrated by 20% alcohol for 20s or 30s,remained satisfactory vitality;While those were infiltrated for 40s or 50s, had been weakened deeply, The longer did the corneal epithelial cells exposed for,the stranger did the corneal epithelial cells were damaged. Conclusion The density of alcoholic solution and the time to be exposed for were very important to laser epithelial keratomileusis（LASEK）.20% alcoholic solution last 20 seconds and especially not more than 30 seconds will be more successful.
