摘  要:目的:探讨信号转导子与转录活化因子1 (STAT1)、核因子κB(NF-κB)p65和凋亡因子caspase 8在胃癌细胞SGC7901中的关系.方法:SGC7901细胞经IFN-γ及STAT1反义寡核苷酸(STAT1ASON)处理后,应用RT-PCR方法检测STALT1,NF-κB p65和caspase 8在mRNA水平的改变.结果:IFN-γ(1000 U)处理SGC7901细胞,在0.5,3和24 h,STAT1和NF-κB p65在mRNA水平出现先下降后上升的变化,二者呈正相关关系(r=0.403,P=0.009);使用不同浓度的STAT1ASON处理SGC7901细胞24 h,在mRNA水平,STAT1和NF-κB p65呈负相关关系(r=-0.556,P=0.015):IFN-γ和不同浓度的STAT1ASON联合作用处理SGC7901细胞24 h,在mRNA水平,STAT1和NF-κB p65呈负相关(r =-0.783,P=0.002),caspase 8呈上升趋势,与STAT1呈负相关关系(r=-0.667,P=0.002),与NF-κ3 p65呈正相关关系(r=0.594,P=0.021).结论:IFN-γ能促进STAT1,NF-κB和caspase 8的表达,STAT1和NF-κB p65在mRNA水平上呈负相关;caspase 8与STAT1呈负相关关系,与NF-κB p65呈正相关关系.AIM: To discuss the relationship between signal transducer and activator of transcription 1 (STAT1), nuclear factor-κB (NF-κB) p65 and apoptotic factor casapase 8, to elucidate the possible mechanism of apoptosis in the human gastric cancer cell line SGC7901. METHODS: SGC7901 cells were treated with IFN-γ and STAT1 antisense oligonucleotides (ASONs). The mRNA expression for STAT1, NF- κB p65 and caspase 8 was exanained by reverse transcription polymerase chain reaction (RT-PCR). RESULTS: When SGC7901 cells were treated with IFN-γ 1000 U, at 0.5, 3 and 24 h, the mRNA levels for STAT1 and NF-κB p65 expression were up-regulated; there was also a positive cor-relation between STAT1 and NF-κB p65 expression (r = 0.403, P = 0.009). With STAT1 ASONs, STAT1 expression was down-regulated, while NF-κB p65 expression was up-regulated; in the mRNA levels, there was a negative correlation between STAT1 and NF-κB p65 (r = -0.556, P -- 0.015). There was no significant correlation between caspase 8 and STAT1 or NF-κB p65. When SGC7901 cells were treated with IFN-γ 1000 U and STAT1 ASONs, there was a negative correlation between mRNA levels of STAT1 and NF-κB p65 (r = -0.783, P 〈 0.05). IFN-γ up-regulated caspase 8 expression; there was a negative cor- relation with STAT1 (r = -0.783, P = 0.002), but a positive correlation with NF-κB p65 (r = -0.667, P = 0.002). CONCLUSION: IFN-γ can up-regulate the mRNA levels of STAT1, NF-κB p65 and caspase 8. mRNA levels of STAT1 and NF-κB p65 were negatively correlated. Caspase 8 had a negative correlation with STAT1, but a positive correlation with NF-κB p65.
