摘  要:[目的]用TK6人类淋巴母细胞的TK基因突变试验评价大蒜素的抗诱变性,为大蒜素抗诱变作用及其可能机制的研究提供毒理学依据,并为TK基因突变试验在抗诱变性检测方面的应用及推广积累资料。[方法]设溶剂对照组、5μg/ml MMS组、5μg/ml MMS+大蒜素0.1μg/ml、2.5μg/ml、5μg/ml、10μg/ml试验组、5μg/ml MMS+100μg/ml VitC对照组。分别采用同时处理和后处理两种方法测定PE0、PE3、RS和TFT。[结果]大蒜素在两种处理条件下均能显著降低MMS诱导的TK基因突变频率,并呈较好的量效关系,两种处理条件的作用强度差异无统计学意义。[结论]在本实验条件下,大蒜素可抑制MMS诱导的TK6细胞TK基因突变,有很好的开发应用前景。[Objective] To detect and evaluate the anti-mutagenesis of allicin by TK gene mutation test, so as to provide toxicological evidence for research on anti-mutagenesis of allicin and its possible mechanism, and to offer information for the application and generalization of TK gene mutation test in detection of anti-mutagenesis. [ Methods ] The TK6 human lymphoid cells were respectively exposed to allicin at concentrations of 0.1, 2.5, 5, 10 μg/ml with 5 μg/ml MMS, 0.1% DMSO, 5 μg/ ml MMS and 5 μg/ml MMS+100 μg/ml VitC. The methods of simultaneous treatment and after treatment were used to determine plating efficiency and mutant frequency, including PE0, PE3, and TFT. [Results] Both treatments showed that allicin could significantly reduce the mutant frequency of TK gene induced by MMS, and the total mutation frequency (TMF) decreased with the increase of allicin dose, besides, statistical analysis showed no significant difference between the two methods. [ Cenclusions] Under the present experimental condition, allicin has obvious antimutagenesis on TK gene induced by MMS, and has a good prospect for development and application.
