温郁金对饮用MNNG大鼠胃黏膜增殖的影响

摘　　要：目的:探讨姜科姜黄属植物温郁金水蒸气蒸馏提取液对饮用化学致癌剂(N-甲基-N’-硝基-亚硝基胍,MNNG)的Wistar大鼠胃黏膜PCNA表达的影响。方法:80只Wistar雄性大鼠随机分为A、B、C、D、E5组,每组16只,A组为空白组,饮用纯净水,生理盐水5mL灌胃1次/天;B组为造模组,自由饮用100μg/mL的MNNG溶液,生理盐水5mL灌胃1次/天;C组:MNNG饮用量同B组,塞来昔布10mg/kg剂量5mL灌胃1次/天;D、E组MNNG饮用量同B组,1g/mL和2g/mL的温郁金水蒸气蒸馏提取液5mL灌胃1次/天。B^E组实验初6周予10%Nacl溶液1mL每周灌胃1次。实验时间40周,实验期间死亡、经病理证实有或无胃肿瘤的动物均为有效动物,计入诱癌情况统计。完成实验之动物于第41周处死,取胃黏膜(已致瘤者选取致瘤部位)免疫组化检测PCNA表达,Leica图像分析系统分析结果。结果:PCNA阳性表达主要见于细胞胞核内,C、D、E组大鼠PCNA积分光密度均低于B组,差异有统计学意义(P<0.05),D组与C组比较PCNA积分光密度差异无统计学意义(P>0.05),E组PCNA积分光密度高于D组和E组,差异有统计学意义(P<0.05)。结论:温郁金水蒸气蒸馏提取液可抑制饮用MNNG大鼠胃黏膜PCNA的表达,这可能是其降低胃癌发生率,显示较好胃癌化学预防能力的机制之一。Objective .To examine the effects of PCNA expression by wet distillation extract of radix curcumae in gastric mucosa of breeded MNNG rats. Methods ： Eighty male Wistar rats were divided into five groups randomly, each 16 members. The rats were given water and 5mL physiologic saline daily （ Group A） ; 5mL physiologic saline daily （ Group B） ;10mg/kg celecoxib daily（ Group C）, 5mL of lg/mL extract of radix curcumae daily （ Group D）, 5mL of 2g/mL extract of radix curcumae daily （ Group E） ;The animals in group B to group E drink water containing MNNG （100μg/mL） freely and given 10% Sodium Chloride in the initial six weeks weekly,All animals received treatment for 40 weeks. Dead rats during the experiment were detected by HE pathological section and the results were bring into ultimate analysis of incidence ratio of induced gatric cancer, the others were sacrificed at week 41th, gastric mucosa （when there is tumor choose tumor tissue）were paraffin -embedded and detected by immunohistochemistry. Results ： PCNA mainly express in cell nucleus, IOD is lower in group C, D,E than group B, there is significant differences in statistics （ P 〈 0.05 ） ;there is no significant differences in statistics when compare group D to group C ,but group E IOD expression is higher then grpup D and group C（ P 〈0.05 ）. Conclusion ：The extract of radix curcumae by wet distillation down regulate PCNA expression in gastric mocosa of MNNG breeded rats. which maybe is one of mechanisms of curcumae to reduce incidence ratio of gatric cancer and displays very good potentiality about gastric cancer chemoprevetion.
