幽门螺杆菌对原癌基因c-Fos和c-Jun表达的影响

摘　　要：目的:探讨幽门螺杆菌(Helicobacter pylori,H.pylori)刺激胃癌细胞AGS后,对细胞内原癌基因c-Fos和c-Jun表达的影响。方法:分别将从不同种类(胃炎,溃疡及胃癌)患者分离到的H.pylori与AGS细胞共同培养1 h,用Western blot方法检测细胞内c-Fos和c-Jun基因表达的变化。结果:不同类型的菌株刺激细胞后,c-Fos和c-Jun蛋白的表达变化存在明显差异。溃疡类菌株只促进c-Fos表达的升高,胃炎类菌株只促进c-Jun表达的升高,而胃癌菌株刺激的细胞c-Fos和c-Jun表达均无明显升高。在不同类型菌株混合刺激下,即胃炎和溃疡菌株,及胃炎、溃疡和胃癌菌株3重混合,刺激后的细胞中c-Fos和c-Jun的表达量均升高。结论:不同H.pylori刺激ACTS细胞后原癌基因c-Fos和c-Jun的表达变化有差异,多类型菌株的混合刺激下c-Fos和c-Jun的表达量同时升高。提示多种类型菌株的复合刺激可能更易促进癌变的发生。Objective：To investigate the expression of c-Fos and c-Jun in AGS cells by stimulation of H.pylori.Methods：The gastric cancer AGS cells were treated with the different types of H.pylori（isolated from gastritis,ulcer or gastric cancer）for one hour,respectively.The lysate of the cells were collected to detect the expression of the two genes by Western blot.Results：The expression of c-Fos protein in the cells stimulated by ulcer type and that of c-Jun by gastritis type increased obviously.Furthermore, the expression of proteins of c-Fos and c-Jun by the stimulation of gastritis and ulcer types mixed was more increased than that of single type of H.pylori and so was the expressions of c-Fos and c-Jun by stimulation of the mixture of the three types.However, there were unremarkable expression changes of the two proteins in the cells treated only with the type of gastric cancer.Conclusion： There was difference in the proto-oncogene c-los and c-jun expression in AGS cells stimulated with different types of H.pylori.The expression of the two genes was more sensitive in the AGS cells treated with multi types of H.pylori.It indicated that the cancer occurrence was promotered by mixed infection of multi types of H.pylori.
