摘  要:研究国产洋根中低聚糖的提取方法和最佳提取条件。以产自浙江省台州地区的新鲜洋根块茎为试材,先测定其主要组分含量,之后依次进行水提S、evag法除蛋白、乙醇沉淀除多糖,从而提取其中的低聚糖。选择固液比、浸提温度和浸提时间为因素,以提取率为指标,建立3因素4水平正交实验,研究低聚糖的最佳提取条件。可溶性总糖的干基含量达到92.30%(W/W)。通过正交实验得到最优提取条件为:固液比1∶4,浸提温度70℃,浸提时间60 min,此条件下低聚糖得率为52.94%(W/W)。经HPLC测定,洋根低聚糖提取物中低聚果糖含量为53.70%(W/W),接近以蔗糖为底物果糖转移酶催化反应得到的商品低聚糖浓度。该提取方法简便易行,稍加改进后即可用于工业化生产。The purpose was to study the extraction method and optimum extraction conditions of oligosaccharide from Chinese Somallanthus sonchifolius. With fresh S. sonchifolius tubers produced from Taizhou district of Zhejiang province as tested materials, the main components content was detected and then the oligosaceharide was extracted from the main components extract by water extraction, seavenging protein by Sevag method and polysaccharide by ethanol precipitation. With ratio of solid to liquid, extraction temperature and extraction time as factors and extraction rate as index, the optimum extraction conditions of oligosaccharide was studied through establishing 3 factor and 4-level orthogonal experiment. The dry matrix content of total soluble sugar reached 92.30%(W/W).The optimum extraction conditions obtained through orthogonal experiment were as follows: ratio of solid to liquid of 1:4, extraction temperature of 70 ℃ and extraction time of 60 min. The extraction rate of oligosaecharides under the conditions was 52.94% (W/W). HPLC determination showed that the fructooligosaccharide content in S. sonchifolius oligosaccharides extract was 53.72% (W/W), which was close to commercial fructcoligosaccharide concentration obtained by fructosyltransferase catalytic reaction with sucrose as substrate. The extraction method was simple and convenient in operation and could be used in industrial production after it was improved further.
