摘  要:用不同浓度刺五加提取物培养RAW264.7巨噬细胞,采用LPS与IFN-γ作为诱导剂,37℃培养24h,Griess试剂检测培养上清液中的NO含量,ELISA法检测培养上清液中的TNF-α含量。结果表明:刺五加提取物在浓度10~1000mg/L内对RAW264.7巨噬细胞分泌的NO具有极显著抑制作用(P<0.01),且呈剂量和时间依赖关系;刺五加提取物对TNF-α的分泌没有影响。这表明刺五加提取物可以显著抑制RAW264.7巨噬细胞产生的NO,而对肿瘤坏死因子-α的产生无明显抑制作用。[Objective] The paper was conducted to investigate the effect of Acanthopanax senticosus extracts on the production of nitric oxide (NO) and tumor necrosis factor-α (TNF-α) in RAW264.7 macrophages induced by lipopolysaccharides (LPS) and interferon-gamma (IFN-γ). [Methods] RAW264.7 macrophages were incubated with LPS and IFN-γ in the presence or absence of Acanthopanax senticosus extracts at various concentrations (1,10,100, or 1000 mg/L ) for 24 h at 37 ℃. The contents of nitrite and TNF-α in the medium were determined by Griess Reagent and ELISA, respectively. [Results] Acanthopanax senticosus extracts (10, 100, or 1000 mg/ L) dose-dependently significantly inhibited NO production in RAW264.7 macrophages induced by LPS and IFN-γ. However, exposure of RAW264.7 macrophages to Acanthopanax senticosus extracts did not affect TNF-α production. [Conclusion] Exposure of RAW264.7 macrophages to Acanthopanax senticosus extracts inhibited NO production induced by LPS and IFN-γ, but had no significant inhibitory effect on the production of tumor necrosis factor-α.
