摘 要:目的探讨天花粉蛋白对体内外单纯疱疹病毒-1(HSV-1)DNA复制的抑制作用。方法昆明小鼠40只随机分为模型对照组和天花粉蛋白治疗组,各20只。模型小鼠颅内接种HSV-1建立病毒性脑炎模型。治疗组于接种HSV-1前30min腹腔注射天花粉蛋白注射液;对照组予等量9g/L盐水。2组于接种病毒后12、24、48、96h分别测定脑组织HSV-1DNA复制水平。体外培养神经细胞分为病毒对照组和药物预处理组,接种病毒后12、24、48、96h分别测定各组培养神经细胞HSV-1DNA复制水平。结果天花粉蛋白治疗组于小鼠颅内接种HSV-1后12、24、48、96h脑组织HSV-1DNA复制水平分别为(1535±656、1468±608、1657±643、1425±534)×106拷贝/mL,显著低于模型组(4336±1350、5256±1837、5379±1975、5946±2769)×106拷贝/mL(Pa<0.001)。药物预处理组在12、24、48、96h后培养神经细胞HSV-1DNA复制水平分别为(1775±743、1347±687、1254±543、1276±525)×106拷贝/mL,显著低于病毒对照组(4653±1851,5120±2110,5045±1867,4995±2578)×106拷贝/mL(Pa<0.001)。结论天花粉蛋白对体内外HSV-1DNA复制具有抑制作用。Objective To explore the inhibitive effect of Trichosanthin(TCS) on herpes simplex virus - 1 ( HSV - 1 ) DNA replication in vivo and vitro. Methods Forty mice were randomly assigned into two groups: Model control group ( model group, n = 20 ), TCS - treated group (treated group,n =20). Viral encephalitis was induced by intracranial inoculation of HSV - 1 in the two groups. The TCS - treated group was injected with TCS 30 minutes before HSV - 1 inoculation. The control group was received equal dose 9 g/L saline. The HSV - 1 DNA replication were measured at 12,24,48,96 h after HSV - 1 inoculation, respectively. The culture neuron in vitro were assigned into two groups:HSV - 1 control group ,TCS -pretreated group. The HSV - 1 DNA replication were measured at 12,24,48,96 h after HSV - 1 inoculated into the neuron, respectively. Results The viral DNA replication of brain tissue at 12,24,48,96 h after HSV - 1 intracranial inoculation were markedly lower in the TCS - treated group ( 1 535± 656,1 468±608,1 657±643,1 425 ±534) × 10^6 copy/mL compared with the model group (4 336±1 350,5 256 ±1 837,5 379 ±1 975,5 946±2 769 ) × 10^6 eopy/mL,respectively ( Pa 〈 0. 001 ). The HSV - 1 DNA replication of neuron at 12,24,48,96 h after HSV - 1 inoculation in TCS - pretreated group( 1 775 ±743,1 347±687,1 254±543,1 276 ± 525 ) × 10^6 eopy/mL were markedly lower than those in the HSV - 1 control group(4 653± 1 851,5 120±2 110,5 045±1 867,4 995±2 578)× 10^6 copy/mL, respectively (Pa 〈 0. 001 ). Conehlsion TCS has inhibitive effects on HSV - 1 DNA replication in vivo and vitro.
