摘  要:从3个地区7个土壤样品中筛选高产菊粉酶的黑曲霉菌株,对筛选出的黑曲霉菌株进行了形态鉴定,探讨了温度、pH值对菊粉酶活力的影响,比较了固体和液体两种扩大培养法对黑曲霉菌株产菊粉酶的影响。结果表明:经过初筛和复筛得到A4、A6、A8、A12、A14、A15和A16共7株黑曲霉菌株,菊粉酶活力大于15.0 U/mL,经数码生物摄影显微镜镜检符合黑曲霉的形态特征。A8菌株的菊粉酶酶活最高,达到25.0U/mL,在60℃、pH值5.0的条件下酶活性最大。Aspergillus niger strains of high inulinase activity was screened from 7 soil samples in 3 regions. The effect of temperature and pH on inulinase activity was tested. The production of Aspergillus niger inulinase were cultivated by solid and liquid state fermentations and compared the activities of the two state fermentations. The result showed that:by means of primary screening and double - screening, 7 high inulinase strains were obtained. They are A4, A6, A8, A12, A14, A15 and A16 and the inulinase activities were up to 15.0U/mL. The 7 high inulinase strains were tested by digital microscope of biological photography and the morphologic features accorded with the standards. The strain A8 showed the highest inulinase activity which hits 25.0U/mL The activities of A8 strain could reach to the highest inulinase activity under the conditions of 60 ℃ and pH5. 0.
