摘 要:通过单因素优化实验、Plackett-Burman实验和Box-Benhnken响应面实验对影响丙酮丁醇梭状芽孢杆菌产丁醇能力的主要因素进行优化,得到了回归方程:Y2=8.583367—0.002125×X7—0.4165×X8+0.297567×X9+0.070692×X10-0.550233×X7^2-0.0379×X7×X8—0.001775×X7×X9—0.05375×X7×X10+0.037529×X8^2-0.182675×X8×X9+0.032475×X8×X10+0.126754×X9^0-0.069×X9×X10-0.588208×X10^2对回归方程进行岭嵴分析,得到了丙酮丁醇梭状芽孢杆菌的优化培养条件,即装液量为310mL,种龄为18h,静态培养,接种量为6%,温度为37℃,初始pH值5.5,在此基础上进行7L发酵罐扩大培养,优化前和优化后的分批发酵结果相比,优化后7L罐最佳浓度的产量为10.55g/L,比原产量8.02g/L提高了31.25%,比有关文献报道的产量高出11.64%。The main characters of Clostridium acetobutylicium influencing the production capacity of Butanol were optimized by using single factor optimization experiment and SAS software combined with the Plackett-Burman design and Box-Benhnken design. A regression equation was obtained. Y2=8.582 367-0. 002 125×X7-0.416 5×X8+0.297 567×X9+0.070 692×X10-0.550 233×X7×X7- 0.037 9×X7×X8-0. 001 775×X7×X9-0.053 75×X7×X10+0.037 529×X8×X8-0. 182 675×X8×X9 +0.032 475×X8×X10+0.126 754×X9×X9-0.069×X9×X10-0.588 208×X10×X10. The ridge analysis was then used to determine the optimal levels of the main factors. The results showed that the optimum fermentation conditions were as follows- 310 mL liquid medium in a flask of 500 mL, inoculums time 18 h, static culture, inoculums size 6%, culture temperature 37℃ and the initial pH 5.5. Under the optimized conditions, the concentration of Butanol in 7 L fermentor increased from 8. 01 g/L to 10.55 g/L, which was 31.25% higher than the production of Butanol before optimization and 11.64 % higher than other related report.
