摘 要:目的通过阶段性观察Wistar鼠大肠癌模型中异常腺窝病灶(ACF)的发展情况,评价ACF与肿瘤的相关性。方法40只Wistar大白鼠给予二甲肼(DMH)皮下注射(20mg/kg,每周1次,连续18周),分为3组,于第8、16和24周分批处死,将美蓝染色后的大肠组织在实体镜下观察。结果ACF均分布于中段及远端大肠(大部分位距盲肠50%~80%处),共发现肿瘤10个,其中3个肿瘤为低分化黏液腺癌,均分布在近端结肠(距盲肠10%~20%)。另外7个肿瘤中有2个为腺瘤,5个为高分化腺癌,均分布在距盲肠50%~80%处。结论ACF与腺瘤癌变途径相关,而非低分化黏液腺癌的中间途径。[Objective] To evaluate the relationship between ACF and tumorigenesis by observing the development from ACF to tumor in DMH-treated rats. [Methods] 40 Wistar rats (s.c.DMH 15 mg/kg bwx18) were killed in different time point at weeks 8,16 and 24, and the colons stained with methylene blue were transilluminated in the stereomicroscopy. [Result] The distribution of most ACFs was between proximal 50%-80% of the colon. In total, 10 tumors developed in 10 rats, and three were seen in proximal 10% and 20%. Histology showed that both of them were poorly differentiated mucin-secreting carcinomas. Seven tumors were found in the distal colon beyond the 50% point. Histology showed two adenomas and five well-differentiated adenocarcinomas. [Conclusion] ACF is an intermediate stage only for the adenoma and the better-differentiated tumors, not the poorly differentiated mucin-secreting carcinomas.
