摘 要:试验以玉米芯粉为唯一碳源,通过富集培养定向筛选技术,从土壤中分离获得一株产木聚糖酶较高的霉菌菌株X-1,经初步液体发酵木聚糖酶活高达724.63U。依据《真菌鉴定手册》初步鉴定该菌株为黑曲霉(Aspergillus nigeer)。同时对该菌株所产木聚糖酶的酶学性质进行了研究,结果表明:该木聚糖酶的最适作用温度为45℃,最适pH值为6.0;在70℃保温30min后酶活力仍为90%以上;在pH3.0—7.0范围内稳定性较好,酶活仍为90%以上。A mold X - 1 strain with high productivity of xylanase was isolated from soil by enrichment culture and orientational screening with maize cob meal as the only carbon source. The xylanase activity of the cultured filtrate came up to 724.63 IU after primarily fermented. It was identified as Aspergillus nigeer. Further research of its enzymology properties showed that, the xylanase was optimally active at 45 ℃and pH 6.0. It was thermostable at 70 ℃ and comparatively stable in the condition of pH 3.0 - 7.0.
