罗格列酮对油酸诱导慢性胰腺炎大鼠肝脏细胞GLUT2mRNA表达的影响

摘  要:目的:观察葡萄糖载体2(GLUT2)在油酸诱导慢性胰腺炎(CP)大鼠肝脏细胞的表达,以及罗格列酮对其mR-NA表达的影响,以进一步探讨罗格列酮防治继发性糖尿病的可能机制。方法:将用油酸诱导的CP大鼠分为模型组、对照组、治疗组,以罗格列酮灌胃治疗6周。RT-PCR法检测各组肝细胞GLUT2 mRNA表达量。结果:模型组GLUT2mRNA表达较对照组明显降低(P<0.05),使用罗格列酮治疗6周后GLUT2 mRNA的表达较模型组明显升高(P<0.05)。结论:慢性胰腺炎可于转录水平阻碍GLUT2的基因表达,这可能是慢性胰腺炎时GLUT2影响肝脏葡萄糖代谢机制之一,罗格列酮在防治继发性胰源性糖尿病动物模型方面具有一定治疗作用。Objective: To observe the expression of glucose transporters 2 (GLUT2) and the effect of rosiglitazone on its mRNA expression and so as to discuss preventing mechanism of rosiglitazone on pancreatogenic diabetes. Methods: The experimental model of chronic pancreatitis rat was induced by oleic acid.Rosiglitazone was feed for 6 weeks. Expression of GLUT2 mRNA was observed by QRT-PCR. Results: The model rats showed remarkable depression of GLUT2 mRNA expression, while rats treated by rosiglitazone demonstrated up-regulation. Conclusion: Chronic pancreatitis can down regulate the expression of glucose transporters 2 on mRNA level, which may be one of the mechanism of GLUT2 on the regulation of glucosemetabolism, rosiglitazone has therapeutic effect on this model to some degree.
