谷物不溶性膳食纤维测定改进方法研究

摘　　要：通过对国家标准GB/T 9822—1988《谷物不溶性膳食纤维的测定方法》的改进研究结果表明:谷物样品先糊化、酶解再用中性洗涤剂处理,比原标准操作中样品先用中性洗涤剂处理再用酶解,解决了过滤困难的问题。其糊化、酶解最佳条件为:样品加水量45 mL,糊化20 min,加2.5%α—淀粉酶溶液5 mL,37±2℃恒温水浴或培养箱保温90 min。GB/T method 9822-1988（determination of insoluble dietary fiber in cereals） was researched. In the method the samples was firstly treated with neutral detergent solution, then enzyme digestion. In the improved method the grain was pasted granule firstly,then enzyme digestion,after that treated with neutral detergent solution. The results showed that the improved method can solve the problem of filtration. Its best condition：adding 45 ml water,pasted granule 20min,adding 2.5%α-amylase 5ml, 37±2℃ constant temperature water bath or keeping temperature for 90 min in incubator.
