摘  要:目的:研究香草醛缩氨基均三唑席夫碱对人肝癌细胞SMMC-7721的诱导分化作用。方法:用不同剂量的3-吡啶-3-基-4-[(4-羟基-3-甲氧基-苯亚甲基)氨基]-5-甲硫基-l,2,4-三唑(LH-37)与SMMC-7721细胞共同培养,以MTF法检测细胞增殖和LH-37对细胞增殖的抑制作用;镜下观察细胞形态改变;采用酶联免疫法检测细胞甲胎蛋白含量,金氏法检测细胞碱性磷酸酶活性的变化。结果:LH-37在1×10^-5-1×10^-3mol/L的浓度范围均能抑制细胞增殖且抑制率随浓度的的增加而增高(P〈0.05或P〈0.01),细胞形态和结构向正常肝细胞方向逆转,1×10^-5mol/L和1×10^-6mol/L的LH-37均可使细胞甲胎蛋白含量明显下降,碱性磷酸酶活性明显升高。结论:香草醛缩氨基均三唑席夫碱对人肝癌SMMC-7721细胞具有抗增殖、促分化作用。Objective: To study the effect of 3 - pyridin - 3 - yl - 4 - [ ( 4 - hydroxy - 3 - methoxy - benzylidene) amino] -5 - methylsulfanyl -4H - [ 1, 2, 4] triazole ( LH - 37) on induction of differentiation of hepatocarcinoma SMMC - 7721 cells. Methods: SMMC -7721 cells were cultured in RPMI - 1640 and treated by LH -37 at different dose. The proliferation of the cells was examined by MTT assay. Cell morphology was observed by a light microscopy. ELI SA method was used to detect the changes of alpha fetoprotein( α - FP) contents of the cells. The specific activities of alkaline phosphatase(ALP) were assayed by ALP kit based on King' S method. Results: The cell proliferation was inhibited by LH -37 at 1 × 10^-5 - 1 × 10^-3mol/L in dose dependent manners. 1 × 10^-5mol/L LH -37 induced the mature and normality of morphology of SMMC-772 1 cells. After treated with 1 × 10^-5mol/L or 1 × 10 ^-6mol/L LH-37, the contents of α- FP was significantly reduced and the activities of ALP were remarkably declined( P 〈 0.0 1). Conclusion: LH - 37 may inhibit proliferation and induce differentiation of human hepatocarcinoma SMMC -772 1 cells.
