摘  要:目的探讨用于脑胶质瘤间质内化疗的BCNU-PLGA缓释微球稳定性的影响因素。方法以溶剂挥发萃取法制备BCNU-PLGA缓释微球,HPLC测定微球内成分,并与空白PLGA微球比较。结果微球冷藏贮存6个月后,外观形态无明显变化,以高效液相色谱方法测定微球溶解后BCNU色谱峰,发现与新鲜配制的BCNU标准品出峰时间和形态一致,微球溶解后BCNU色谱峰表现为单一尖锐峰形,表明药物在微球内部保持结构和成分的稳定。结论BCNU-PLGA缓释微球具有良好的稳定性,可以满足间质内缓释化疗的需要。Objective To explore the influential factors of stability to BCNU-PLGA sustained release microspheres used for interstitial chemotherapy to brain glioma. Methods BCNU-loaded PLGA microspheres are fabricated and prepared by solvent evaporation and extraction, components are determined by HPLC, and compared with blank loaded microspheres. Results Morphous without significant difference, chromatographic peak are identified between solved BCNU and fresh prepared BCNU standard solution, the chromatographic curve after solved is signal cut-edge one, indicated that the drug remaind stable inside the microspheres. Conclusions Stability of BCNU-PLGA sustained release microspheres is well enough to satisfactory the requisition of interstitial sustained release chemotherapy.
