牛磺酸对S_(180)移植瘤小鼠抗氧化作用

摘  要:目的观察牛磺酸对S180移植瘤小鼠抗氧化能力的影响。方法60只昆明小鼠随机分为牛磺酸低(A)、中(B)、高(C)剂量组(分别在饮水中添加0.5%,1.0%,1.5%的牛磺酸),环磷酰胺阳性对照组(D),肿瘤阴性对照组(E)。各组小鼠左前肢腋窝皮下均接种S180细胞悬液0.2 ml。24 h后,阳性对照组腹腔注射环磷酰胺20 mg/(kg.bw)。1周后处死小鼠,测定血浆、肝组织中超氧化物歧化酶(SOD)和谷胱甘肽过氧化物酶(GSH-Px)的活力、丙二醛(MDA)含量;采用H2O2诱发氧化溶血实验检测红细胞的溶血度;采用单细胞凝胶电泳(SCGE)技术测定血淋巴细胞的DNA断裂情况。结果(1)与阴性对照组比较,牛磺酸各剂量组小鼠血浆中MDA含量降低(P<0.01),血浆、肝脏的SOD和GSH-Px活力增强(P<0.05或P<0.01)。(2)牛磺酸低、高剂量组溶血度与阴性对照组比较差异有统计学意义(P<0.05)。(3)用5,10和25μmol/L H2O2处理血淋巴细胞后,牛磺酸各剂量组的DNA损伤较阴性对照组低,其中10μmol/L H2O2处理的牛磺酸中、高剂量组和25μmol/L,H2O2处理的牛磺酸高剂量组的DNA损伤与阴性对照组比较差异有统计学意义(P<0.05或P<0.01)。结论牛磺酸能提高S180移植瘤小鼠的抗氧化能力,可能与提高机体抗氧化酶活力,减少脂质过氧化物含量,保护红细胞膜,拮抗DNA氧化损伤有关。Objective To investigate the antioxldant activities of tanrine on sarcoma 180($180) bearing mites. Methods Sixty Kunming mites were divided randomly into five groups. Groups A, B, C were drank with 0.5 %, 1.0 %, 1.5 % taufine respectively;group D was cyclophosphamide positive control; group E was the tumor negative control. Then mites were vaccinated with 0.2 ml S180 cell suspension in left axillas. 24 hours later, the positive control was injected by cyclophosphamide 20 mg/(kg, bw). After 1 week, the content of maleic dialdehyde(MDA) and the activity of superoxide dismutase (SOD) and glutathione peroxidase (GSH- Px) in plasma and liver were detected with test kit respectively. Erythrocytic haemolysis rate induced by H2O2 was studied. Lymphocyte were separated for determination of DNA damage by using SCGE. Results Compared with negative control, the activity of SOD and GSH - Px in plasma and liver of tanrine groups increased( P 〈 0.05 or P 〈 0.01 ) and the content of MDA in plasma decreased ( P 〈 0.01 ) significantly. Erythrocytic haemolysis rate of 0.5 % and 1.5 % tanrine groups were markedly lower than the negative control ( P 〈 0.05). There was a decreasing tendency of the oxidative DNA damage on peripheral lymphoeytes induced by 5, 10 and 25μmol/L H2O2. The DNA damage was obviously lower than the negative control in 1.0 % and 1.5 % tanrine groups induced by 10μmol/L H2O2 and in 1.5 % tanrine group induced by 25μmol/L H2O2(P〈 0.05 or P 〈 0.01 ). Conclusion Taurine can improve antioxidative effect of S180, bearing mites, which may be associated with its effects of increasing antioxidase activity and reducing lipid peroxidation and protecting the membrane of erythroeytic and resisting oxidative DNA damage.
