摘 要:目的探讨外源性嘌呤核苷酸对吗啡致神经细胞核苷酸分解代谢增强的影响。方法应用MTT比色法检测给予吗啡同时给予嘌呤核苷酸(AMP和GMP及ATP和GTP)对PC12细胞存活率的影响;RT-PCR法检测腺苷脱氨酶(ADA)和黄嘌呤氧化酶(XO)mRNA表达。结果同时给予吗啡和嘌呤核苷酸可改善吗啡对PC12细胞增殖的抑制作用。给予吗啡并补充AMP和GMP使得ADA mRNA表达与对照组及吗啡组比较均无差异,XO mRNA表达低于吗啡组(P<0.01);给予吗啡及ATP和GTP使得ADA mRNA表达仍高于对照组(P<0.05),XO mRNA表达低于吗啡组(P<0.05)。结论补充嘌呤核苷酸可改善吗啡对PC12细胞增殖的抑制作用,减弱吗啡所致的PC12细胞ADA和XO基因表达升高。Objective To explore the effect of exogenous purine nucleotide on the augmented atabolism of neurocyte nucleotide induced by morphine. Methods The effect of morphine and purine nucleotide (AMP, GMP, ATP and GTP) interventions on PC12 cell survival rate were determined by MTr chromatometry. The mRNA expressions of adenosine deaminase (ADA) and xanthine oxidase (XO) were detected by RT-PCR. Results Morphine and purine nucleotide interventions improved the inhibition of morphine on PC12 cell proliferation. In PC12 cells with morphine + AMP + GMP, ADA mRNA expression did no differ from that in control and morphine groups, XO mRNA expression was lower than that in morphine group (P 〈0. 01 ). In PC12 cells with morphine + ATP + GTP, ADA mRNA expression was higher than that in control group (P 〈 0. 05 ) , XO mRNA expression was lower than that in morphine group ( P 〈 0. 05 ). Conclusions Purine nucleotide supplement could improve the inhibition on PC12 cell proliferation induced by morphine and weaken the increased ADA and XO gene expressions of PC12 cell induced by morphine.
