摘 要:研究了木质层孔菌不同发酵条件对木聚糖酶、β-葡聚糖酶性能的影响,并对其酶作用特性进行了研究结果表明:木质层孔菌产木聚糖酶最适碳源为麸皮,氮源为蛋白胨,最适发酵温度为25℃;而产β-葡聚糖酶最适碳源为木糖,氮源为酵母膏,最适发酵温度为28℃。就产酶时间而言,在发酵前60 h菌株均不产生β-葡聚糖酶和木聚糖酶,在72 h才有微量的酶产生,到192~240 h两种酶均达到产酶高峰,并以216h产酶最高;两种酶的最佳缓冲液均为柠檬酸缓冲液,木聚糖酶的最适作用pH值5.0,最适反应温度为50℃,而β-葡聚糖酶最适作用pH值4.6,最适反应温度为60℃。Different fermentation conditions which affected the performance of xylanase and β -glucanase of Fomes lignosns (Brek) Ccoke and characteristic of its enzyme were studied. The results showed that the optimal carbon source for Fomes lignosns (Brek) Ccoke to produce xylanase was wheat bran, the optimal nitrogen source was yeast extract, the optimal ferment temperature was 25℃. The optimal carbon source for Fomes lignosns (Brek) Ccoke to produce β-glucanase was xylose, the optimal nitrogen source was yeast extract, the optimal ferment temperature was 28 ℃. For producing enzyme time, the Fomes lignosns (Brek) Ccoke didn't produce xylanase and β-glucanase until it fermented for 60h. Minim enzyme was produced when strain 5.132 fermented for 72h and enzyme producing fastigium is 192h -240h. Maximal enzyme was produced at 216h. The optimal buffer for xylanase and β-glucanase was citric acid buffer, the optimal pH for xylanase was 5.0, the optimal reaction temperature was 50℃, the optimal pH for β-glucanase was 4.6, the optimal reaction temperature was 60℃.
