摘  要:目的探讨尼卡地平对脆性X综合征动物模型(FMR1基因敲除小鼠)的治疗效果及行为学评价。方法成年野生型和FMR1基因敲除鼠(各8只)随机分为尼卡地平治疗组、生理盐水对照组(各4只)。使用Morris水迷宫实验评价脆性X综合征模型小鼠尼卡地平治疗30d后对学习记忆的影响,并应用旷场行为实验和孔板探究实验研究治疗前后精神情绪与自主行为的改变。结果Morris水迷宫实验表明,与野生型小鼠比较,FMR1基因敲除小鼠找到迷宫平台潜伏时间延长,通过平台区域的次数下降;旷场行为实验与孔板探究实验结果显示,模型小鼠探索性行为、自主活动明显增加。尼卡地平治疗后,与模型小鼠对照组比较,模型小鼠治疗组Morris水迷宫实验未见明显改变,而自发活动显著减少(P〈0.05)。结论FMR1基因敲除小鼠存在空间学习记忆能力障碍,自主活动亢进,尼卡地平对学习记忆的改善无明显作用,但可减少其自主活动的高兴奋性,具有一定的治疗效果。Objective To study the curative effects of nicardipine on fragile X syndrome animal models (FMR1 gene knockout mice) by observing their behavior change. Methods The experimental mice (adult wild-type and FMR1 gene knockout mice) were randomly divided into 4 groups: control groups (WT & KO) and treatment groups (WT & KO). The treatment groups were injected with nicardipine for 30 d and the control groups with normal saline instead. Morris water maze experiment was used to evaluate the spatial learning and memory abilities of mice after the treatment of nicardipine. And open field and hole-board exploration tests were applied to evaluate the change in moods and spontaneous activities of mice. Results In Morris water maze experiment, the escape latency was longer and the frequency of crossing the platform was less in KO mice than in WT mice; In open field and hole-board exploration tests, the KO mice displayed a marked increase of exploration behaviors and spontaneous activities compared with those of WT mice. The group of nicardipine treated KO mice made no difference with the group of control KO mice in the Morris water maze experiments, but had a significant decrease in spontaneous activities in open field test (P〈0.05). Conclusion FMR1 knockout mice have a defect in spatial learning and memory abilities, and excessive spontaneous activities. Nicardipine has no significant effect on learning and memory abilities of KO mice, but it can decrease excessive locomotor activities and relieve anxiety in KO mice.
