摘  要:以轮梗霉紫外诱变突变株A1.13为出发菌株,制备其原生质体并连续采用紫外线、微波及硫酸二乙酯作为诱变剂进行多轮诱变,通过15℃低温和0.12g/ml乙酰水杨酸初筛及摇瓶发酵复筛,得到花生四烯酸稳定提高的突变株MW40+DES20II-4,其花生四烯酸占总脂肪酸的含量从出发株的8.034%提高到15.098%。采用5L全自动发酵罐发酵,其生物量、油脂产量和花生四烯酸产量分别达到12.252、4.307g/L和604.557mg/L,分别比对照提高了24.96%、56.39%和193.924%。通过突变株的脂肪酸组分分析,认为多轮诱变对于脂肪酸生产菌是有效的育种手段。Using Diasporangium sp. Al.13 as original strain, the protoplast mutagenesis by UV, microwave and DES were carded on in order to enhance the arachidonic acid yield. The mutant MW40+DES20 II-4 was selected through low temperature (15℃) and aspirin (0.12 kg/ml) as inhibitors with the result of the AA content in total fatty acids increasing from 8.034% to 15.098%. Fermentation experiment was carried on 5 L fermentation equipment. Results showed that the biomass, arachidonic acid yield and fatty acid yield reaches 12.252 g/L, 4.307 g/L and 604.557 mg/L respectively increasing 24.96%, 56.39% and 193.924% than the initial strain. It is thought that it is an effective breeding means for fungus producing fatty acids through analysis the fatty acid component of the mutants.
